Donald Edward Newhouse was born on August 5, 1929, in New York City. He is the son of Samuel Irving Newhouse Sr., who founded Advance Publications in 1922, and Mitzi Epstein Newhouse, an arts patron and philanthropist. Newhouse attended Syracuse University but left after his freshman year to enter the family business around 1949. His upbringing provided early exposure to the practicalities of newspaper management and journalistic operations.

He and his late wife, Susan, also donated to Wellesley College, establishing the Susan and Donald Newhouse Center for the Humanities.
