Jon Lloyd Stryker was born in Kalamazoo, Michigan, in 1958. He is the grandson of Homer Hartman Stryker, who founded the medical supply company Stryker Corporation. His father, Lee Stryker, died in a plane crash in 1976. Stryker earned a Bachelor of Arts degree in biology from Kalamazoo College in 1982 and a Master of Architecture degree from the University of California, Berkeley. He is a registered architect in Michigan.

Stryker's wealth comes from his family's stake in Stryker Corporation, a global medical technology company. While he does not hold an operational role in the company, he has a significant inherited interest.

Stryker is the founder and president of the Arcus Foundation, a private international philanthropic organization that primarily supports great ape conservation efforts and LGBT causes, as well as other social justice endeavors. The Arcus Foundation has awarded over $500 million in grants. He has donated more than $700 million to various causes. In 2021, Jon and his husband, Slobodan Randjelović, gave $15 million to the ACLU Foundation to focus on LGBTQ rights and HIV issues.
